AOCs are moving into clinical development across a growing range of therapeutic areas, from neuromuscular and rare genetic disease to CNS disorders and immuno-oncology. This blog reviews named clinical programmes, including Avidity's TfR1-targeting siRNA conjugate in myotonic dystrophy and Tallac's CD22-targeting immunostimulatory conjugate in oncology, and sets AOCs against the other delivery platforms competing for the same problem: local administration, chemical modification, LNPs, viral vectors and GalNAc conjugates. The comparison makes the case for where AOCs offer a genuine advantage, particularly in extending targeted delivery beyond the liver.
